Scientists Discover a Vagus Nerve Signal That Links Food Directly to Memory Formation - ScienceAlert

ScienceAlert reports that a new study in Nature Communications has identified a gut-to-brain signaling pathway that may help explain why meals can be closely tied to memory formation. The work, led by University of Southern California biologist Scott Kanoski, examined rats and found that after nutrient intake, signals traveling along the vagus nerve from the gut influenced neurons in the medial septum, a brain area that helps regulate hippocampal activity.

According to the report, the researchers focused on acetylcholine, a neurotransmitter released by medial septum neurons that is known to be involved in encoding memories in the hippocampus. Using fiber photometry, the team monitored brain-cell activity while rats consumed different diets. Rats given fat or sugar showed stronger memory-related brain activity than animals given sweet-tasting but low- or no-calorie solutions, suggesting that calories or nutrients, rather than taste or eating itself, drove the observed signaling effect.

The study also found limits to the apparent benefit. ScienceAlert says rats exposed early in life to a high-fat, high-sugar “junk food” diet later showed a weakened acetylcholine signaling system and performed worse on memory tasks, even after returning to healthier food. The researchers reported that disrupting vagal input or deleting relevant acetylcholine neurons eliminated the food-linked effect.

The findings are preliminary because they come from rat experiments and, as ScienceAlert notes, only male rats were studied. Still, the work may be relevant to future research on human gut-brain signaling, including interest in vagus nerve stimulation. The article notes that acetylcholine disruption is an early neurochemical feature of Alzheimer’s disease, making the pathway a possible target for further investigation.
