Fox News reported that an experimental pancreatic cancer prevention vaccine from Johns Hopkins School of Medicine showed encouraging results in an early human study involving people considered at elevated risk, but who had not been diagnosed with the disease. The phase 1 trial, published in Cancer Discovery, tested mKRAS-VAX in 20 participants with inherited risk for pancreatic ductal adenocarcinoma and imaging abnormalities, most often pancreatic cysts.

According to the report, participants received four initial injections during weeks one, three and five, followed by a booster at week 13. The vaccine was designed to target six common KRAS mutations, a gene pathway linked to most pancreatic ductal adenocarcinomas. Researchers said 18 of 20 participants developed immune responses directed at mutant KRAS, and immune activity rose on average about 18-fold, though individual responses differed.

Fox News cited Johns Hopkins researchers as saying the vaccine appeared safe in this small trial, with side effects limited to injection-site reactions and short-lived flu-like symptoms. After a median follow-up of about 16.5 months, no participants had developed pancreatic cancer, and 37.5% had shrinkage or disappearance of the cysts associated with their increased risk.

The findings matter because pancreatic ductal adenocarcinoma is often aggressive and difficult to detect early. However, the researchers emphasized that this was a small phase 1 study designed mainly to assess safety, not to prove prevention or clinical effectiveness. They said larger studies are needed to determine whether the cyst changes and cancer-free follow-up were caused by the vaccine and to identify the best targets and timing for vaccination in high-risk groups.
