Medical Xpress reported on July 23, 2026, that a UCLA Health-led mouse study found inflammation during pregnancy can produce autism-like brain and behavioral changes in offspring, and that some of those effects could be eased rapidly but briefly in adult mice after one dose of rapamycin. The study was published in Nature Communications and examined a maternal inflammation model, not human patients.

According to the report, pregnant mice were exposed to a mild inflammatory stimulus early in gestation. Their offspring later showed persistent inflammation, slight brain overgrowth, altered mTOR pathway signaling, disrupted brain network activity and behaviors associated with autism, including repetitive actions and sensory over-responsivity. When adult offspring received rapamycin, researchers observed improvements within about two hours in neuron activity, seizure susceptibility, brain connectivity patterns and some behaviors.

The UCLA team said the speed of the response suggests the drug was not repairing underlying structural brain differences, which would typically take longer, but was instead changing functional brain activity. Researchers also reported rapid shifts in gene activity related to autism, epilepsy and ion channel function, especially in excitatory neurons, pointing to brain-cell excitability and network balance as possible future treatment targets.

Medical Xpress emphasized that the researchers did not present rapamycin as a treatment for autism symptoms in people. The effects in mice were temporary, repeated daily dosing led to tolerance over several weeks, and rapamycin can have toxic effects. The findings matter mainly because they suggest adult brain circuits may remain more functionally adjustable than previously assumed, offering directions for future research beyond this specific drug.
